AstraZeneca - AZD7442 long-acting antibody combination approved for prevention and treatment of COVID-19 in Japan



First global approval for AZD7442 as a COVID-19 treatment

PROVENT Phase III prevention trial showed reduced risk of developing symptomatic COVID-19

TACKLE Phase III treatment trial showed reduced risk of severe COVID-19 or death in high-risk patients


AstraZeneca's AZD7442 (tixagevimab and cilgavimab), a long-acting antibody combination, has been approved in Japan for both prevention (pre-exposure prophylaxis) and treatment of symptomatic disease caused by SARS-CoV-2 infection. The decision marks the first global marketing approval for AZD7442 as a treatment for COVID-19.


In prevention, Japan’s Ministry of Health, Labour and Welfare (MHLW) granted AZD7442 Special Approval for Emergency for adults and adolescents (12 years of age and older weighing at least 40kg). AZD7442 is approved for use in those whom SARS-CoV-2 vaccination is not recommended and who may have an inadequate response to a COVID-19 vaccine due to immunodeficiencies. Recipients of AZD7442 for prevention should not be currently infected with or have had recent known exposure to a person infected with SARS- CoV-2.


In treatment, AZD7442 is approved for adults and adolescents (12 years of age and older weighing at least 40kg) with risk factors for severe SARS-CoV-2 infection who do not require supplemental oxygen.


Kazuhiro Tateda, M.D. Ph.D., Professor, Department of Microbiology and Infectious Disease, Toho University, Tokyo, Japan, said: “COVID-19 continues to have a significant impact on our daily lives in Japan. Many people, including older adults, patients with comorbidities, and immunocompromised patients, remain at risk for poor outcomes from severe COVID-19. AZD7442 will be a much-needed new option, offering long-term protection for those who do not achieve an adequate immune response after vaccination and helping prevent severe disease and death in those who do become infected.”


Itaru Matsumura, M.D., Ph.D. Professor & Chairman, Department of Hematology & Rheumatology, Kindai University Faculty of Medicine, Otsuka, Japan, said: “Despite the progress of vaccinations and stringent safety precautions, there are very large number of new infections in Japan. The approval of AZD7442 is expected to provide a non-vaccine prophylactic option for those who cannot expect a full immune response from COVID-19 vaccination, such as patients with blood cancers.”


Mene Pangalos, Executive Vice President, BioPharmaceuticals R&D, AstraZeneca, said: The approvals of AZD7442 in Japan represent an important milestone in our ongoing efforts to help combat COVID-19 on all fronts. AZD7442 is now the only long-acting antibody combination authorised for both COVID-19 prevention and treatment, allowing us to help protect even more vulnerable patients such as the immunocompromised from this devastating disease.”


The Japanese government has agreed to purchase 300,000 units of AZD7442 (150mg each of tixagevimab and cilgavimab) and AstraZeneca is working with the government and partners to make first doses available as soon as possible.


The approvals were based on efficacy and safety data from the AZD7442 clinical development programme, including the PROVENT Phase III pre-exposure prophylaxis trial, the TACKLE Phase III outpatient treatment trial, and Phase I trials, including in Japan. In PROVENT, a 300mg intramuscular (IM) dose of AZD7442 significantly reduced the risk of developing symptomatic COVID-19 by 77% (95% confidence interval (CI): 46, 90; p<0.001) COVID-19 compared to placebo at the primary analysis.An 83% (95% CI: 66, 91) relative risk reduction was shown at a six-month median follow-up analysis, with protection from the virus lasting six months.1


In TACKLE, a 600mg IM dose of AZD7442 significantly reduced the relative risk of progressing to severe COVID-19 or death (from any cause) by 50% (95% confidence interval [CI] 15, 71; p=0.010) through day 29 compared to placebo in non-hospitalised patients with mild-to-moderate COVID-19 who were symptomatic for seven days or less, the trial’sprimary endpoint.In pre-specified analyses of participants who received treatment within three days of symptom onset, AZD7442 reduced the risk of developing severe COVID-19 or death (from any cause) by 88% compared to placebo (95% CI 9, 98), and the risk reduction was 67% (95% CI 31, 84) when participants received AZD7442 within five days of symptom onset.2


AZD7442 was generally well-tolerated in the trials.1,2

The recommended dose for prevention of symptomatic disease caused by SARS-CoV-2 infection in Japan is 150mg of tixagevimab and 150mg of cilgavimab, administered as separate sequential IM injections. Depending on the prevalence of SARS-CoV-2 variants, 300mg of tixagevimab and 300mg of cilgavimab may be administered for prevention. The recommended dose for treatment of COVID-19 is 300mg of tixagevimab and 300mg of cilgavimab, administered as separate sequential IM injections.


AZD7442 has been shown to retain in vitro neutralisation activity against the main Omicron variants currently circulating globally, including BA.5 and BA.2.3,4


AZD7442 is also authorised for use for pre-exposure prophylaxis (prevention) of COVID-19 in the US (emergency use), EU and many other countries. Regulatory submissions are progressing for both prevention and treatment indications around the world.

City to build city college beside CDRRMD new building in Zone 8, Carmen, CDO


The City Government of Cagayan de Oro is set to construct the building for the city college in Zone 8, Barangay Carmen, beside the soon-to-rise CDRRMD Disaster Coordination Center next year.


However, Mayor Rolando "Klarex" Uy said the city college will begin offering its courses during this school year's second semester.


"Makasiguro ko labing dugay sunod tuig pero paningkamutan nako sa second semester karong tuiga. Silbi karong school year second semester, makasugod ta sa city college, bisa’g walay building dinhi," Mayor Klarex Uy said.


"Nagpasalamat ko kay nagsaad pud ang DepEd nga aduna sila’y building nga pahulaman, nga makasugod ta sa atong city college," he added.


The bulk of the possible students the city college will cater consist of the out-of-school youths and those who are currently employed but have not finished any college degree yet, according to Cagayan de Oro Technical Vocational Institute (CDO TVI) School Vocational Administrator Maria Victoria Trinidad.


At present, the technical working group for the establishment of the city college is conducting its feasibility study.


It will determine the kind of programs the city college will offer, the kind of faculty it will have, and the extent of the project development plan of the campus.


"We also want to make sure that the one who will be managing it will really bring the city college to its best form and its best service possible," Trinidad said.

Globe scales up network builds to serve remote public schools


Globe continues to scale up its network builds and upgrades, especially in geographically isolated, disadvantaged, and conflict-affected areas to help provide connectivity to 17,000 public schools that still do not have data or internet access.

This was shared by Michael Agustin, Director for Enterprise Sales of Globe, one of the panelists at the Industry Leaders Conference on Connecting Last Mile Schools organized by the United States Agency for International Development (USAID) through All Children Reading Philippines (ACR-Philippines), Better Access and Connectivity (Beacon), and Advancing Basic Education in the Philippines (ABC+).


Agustin talked about closing the digital divide in education through network expansion and inclusive connectivity deployments.


“We recognize that the network infrastructure remains the core foundation for digital inclusion. That is why we continue to work with the government, non-government institutions and the community in addressing the inequalities of the digital divide by democratizing access, enabling platforms, and helping educators embrace technology,” he said.


Agustin noted how Globe earmarked P89 billion this year for its capital expenditures.  For the first half of 2022, it has already spent P22 billion on the construction of 572 new cell towers. An additional 1,700 cell sites are expected to be installed all over the Philippines by yearend to reach more unserved and underserved Filipinos.


At the same time, the company continues to roll out 4G/LTE across its network to further improve customer data experience.  It is also adding more 5G sites for better connectivity.


Globe also plans to build an Information and Communications Technology (ICT) backbone in Mindanao and is now in talks with the Mindanao Development Authority.


It is partnering with regional providers who are present in remote areas and is coordinating with local government units (LGUs) and communities to see how Globe can provide services and improve signal. 


“We're working closely with provinces, cities, and municipalities in identifying the areas because building towers and our network across the country really needs the help of the LGUs and the communities themselves,” Agustin said.


Aside from network expansion, the digital solutions platform is working with the Mindanao Development Authority in mapping out and identifying projects in their roadmap, especially under the people's well-being program. This includes education and health and wellness in different municipalities across the Bangsamoro Autonomous Region in Muslim Mindanao (BARMM). 

 

“Education is very important to Globe. We believe that the students today are the nation builders of tomorrow. To uplift the lives of Filipinos, we help students, teachers, and school administrators overcome challenges by providing digital technology so that learning is continuous no matter what,” said Agustin.


He added: “Technology can level the playing field for all especially those who are at a disadvantage. There's so much to do amidst all these challenges but opportunities abound. We only need to work together as partners and collaborators.”


Globe has embarked on various initiatives to make education more meaningful, effective, and uniform across all schools in the country. It helps transform public high schools into ICT centers of excellence by equipping students and teachers with the knowledge, skills, and tools needed for the future through the Global Filipino Schools (GFS) program.


GFS is part of a long-term partnership between Globe and the Department of Education (DepEd) meant to forward 21st Century Learning principles and ICT integration in public schools nationwide. About 414 schools across all regions in the Philippines, the latest of which are in BARMM, are provided with internet, ICT equipment, and training for teachers. 


Likewise, Globe keeps students safe and molds them into responsible online citizens through the Digital Thumbprint Program (DTP), a campaign to combat fake news, misinformation, and cyberbullying. The program is integrated into DepEd's curriculum, having provided access to 1.9 million students across the country.  About 15,000 teachers and 4,000 schools have participated in the program.


Likewise, the Globe e-library provides access to digital storybooks, learning videos, and other resources for free for K-12 students. Globe and TM users can access the site even without load. 


Globe also conducts free webinars for administrators and teachers in delivering the best hybrid learning experience through eSkwela. One was held in August to discuss education technologies. 


In addition, companies under the Globe Group are doing their share in contributing to quality education and livelihood. 


Edventure, an online learning platform under 917Ventures, provides virtual one-on-one and personalized tutorials for students while offering professional opportunities for Filipino tutors nationwide.

This year’s KUMBIRA will adopt the theme “TUBONG MINDANAW” making known local cuisine and products of the different regions of Mindanao (Regions: IX, X, XI, XII, XIII and BARMM) , as well as promote ingredients indigenous to this part of the Philippines. This year's Kumbira will take place on October 15-17, 2022 at the Limketkai Mall Atrium.
